Installation
During installation, I am asked to select a USB port, but I don't know which one to select.

You can check the port number from Device Manager:
Start-> Settings-> Control Panel-> System-> Hardware-> Device Manager...->
Ports (COM & LPT)
There you should see "Port 1 on Nokia Adapter (COM3)" or similar information.
In this case your cable is shown in COM3 and you should select it from the list during installation.

When installing PC Suite 5.03, I manually selected the USB port where the phone was connected. When running PC Suite 5.03 after installation, the connection does not work.

Make sure that the communication port you specified during Setup is the right one. If you selected the wrong communication port, open the Connection Manager from the Control Panel and click the Advanced button to change the USB port.
